Abstract
One of the most classical and well known in vitro system is the Ames test, making use of a liver homogenate providing the metabolic competence needed for the detection of mutagenic reactive metabolites in Salmonella bacterium. Liver cells were isolated and have been tried over many years to maintain their differentiation status. The application of established cell lines for metabolism related studies was developed. The importance of cytochromes P450 became evident in the meantime, resulting into studies based on individual purified and reconstituted cytochromes P450. Finally, genetically engineered cell systems for metabolism studies were created (Doehmer 1993).
