Abstract

Taxonomy has been one of the major problems in mycorrhizal research ever since the discovery of endomycorrhizal fungi. Identification of individual species is a prerequisite for taxonomy; however, owing to the fact that hyphae of different fungi are often virtually indistinguishable from one another, traditional means of taxonomy still rely mostly on morphological characters of resting spores in soil, such as spore size, shape, wall structure and composition, color, and ornamentation. Unfortunately, some of these characters vary not only with the developmental stages of spores as these are formed, but are also instable after fixation, resulting in difficulties producing viable specimens for future reference.
