Abstract

The protocol described below has been used successfully to localise RNA transcripts for a number of genes expressed in various tissues (leaf, apex, flower, stem, fruit, root) of tomato (Fleming et al. 1993), tobacco (Fleming et al. 1992) and Spirodela (Smart and Fleming 1993). It is adapted from the original method described by Cox and Goldberg (1988). To minimise RNA degradation, aqueous solutions should be autoclaved (when possible), glassware baked, and sterile plasticware used. Follow standard procedures to exclude RNase contamination.
