Abstract
Gene transfer to meristems is an attractive approach to many problems of basic and applied plant science. Microballistics are the methods of choice to transform meristem cells. Shoot apical meristems are small and sensitive and therefore difficult to be handled in conventional ballistic systems. To transform as many cells as possible of a single meristem, we developed a ballistic microtargeting system, which allows the precise delivery of microprojectiles into shoot meristems (Sautter et al. 1991). Here we describe the delivery of the bacterial gene for beta-glucuronidase (Jefferson et al. 1987) into shoot apical meristems of germinating wheat seedlings, and the detection of the gene product by a histochemical assay (Bilang et al. 1993).
