Zusammenfassung
Die Pankreaslipase und ihr Kofaktor, die Kolipase, gelangen über den Pankreassaft ins Duodenum. Während die Lipase fertig synthetisiert bereitgestellt wird, gelangt letztere zunächst als Prokolipase ins Duodenum. Die Prozessierung zur Kolipase setzt ein N-terminales Pentapeptid (Enterostatin) frei, das an der Regulation der Fettaufnahme und des Körpergewichts beteiligt sein soll. Die Lipase ist zur Entfaltung ihrer katalytischen Aktivität auf eine saubere Öl-Wasser-Grenzfläche im Darmlumen angewiesen, für die die grenzflächenaktiven Gallensalze sorgen. Daß trotz Anwesenheit der Gallensalze die Lipase funktionell an die Grenzfläche anbinden kann, verdankt sie der Komplexbildung mit der Kolipase. Ein genauer Einblick in die Struktur des sich ausbildenden Lipase-Kolipase-Komplexes wurde durch die Aufklärung der Tertiärstruktur mit Hilfe der Röntgenstrukturanalyse möglich. Das aktive Zentrum, die katalytische Triade (Asp-His-Ser), liegt in einer hydrophoben Tasche und ist nach außen durch ein Lid verdeckt. Erst durch die Bildung des spezifischen Lipase-Kolipase-Komplexes bringt der Kofaktor die Pankreaslipase in direkten Kontakt zur Öl-Wasser-Grenzfläche, der mit der Öffnung des Lids und somit der Freigabe des katalytischen Zentrums einhergeht. Diese Grenzflächenaktivierung der Pankreaslipase ermöglicht die Lipolyse im Darmlumen.
