Mass Spectrometry in Drug Disposition and Pharmacokinetics

Abstract

An examination of the recent literature suggests that the role of mass spectrometry (MS) in the fields of drug disposition and pharmacokinetics is expanding. This is most likely a consequence of the availability of new instrumentation and ionization techniques that can be used to help solve difficult bioanalytical problems associated with these fields (Burlingame et al. 1990, 1992). This can perhaps be best illustrated in the development of electrospray (ESI) and related atmospheric pressure ionization (API) techniques such as ionspray and atmospheric pressure chemical ionization (APCI) for studies in drug disposition. Over the last 2 years there has been an unprecedented explosion in the use of instrumentation dedicated to Apims (Smith et al. 1990; Fenn et al. 1990; Hamdan and Curcuruto 1991; Huang et al. 1990; Huang and Henion 1991; Bruins 1991; Wachs et al. 1991). Perusal of the literature would suggest that thermospray (TSP) is still the most popular liquid chromatography (LC)/MS technique for structural and quantitative studies (Arpino 1990). However, this is to some extent a reflection of the fact that many APIMS studies have not been reported in full and remain only in abstract form (predominantly in Proceedings of 39th and 40th ASMS Conferences on Mass Spectrometry). With time there is no doubt that the full papers derived from these initial reports will eventually appear. Perhaps another major factor slowing publication is that many studies have been carried out with proprietary compounds and it will take several years for patent clearance to be obtained for the full release of all the findings to take place. The pharmaceutical industry has certainly been at the forefront of implementing API–based methodology and this perhaps reflects the utility of these techniques for studies on drug disposition. Several reviews have appeared recently on the use of MS in drug disposition (Baillie 1992; Garland and Vandenheuvel 1992), pharmacology (Abramson 1990; Fenselau 1992), and biology (Burlingame and McCloskey 1990; Suelter and Watson 1990; McEwen and Larsen 1990; McCloskey 1991; Gross 1992; Stroh and Rinehart 1992).
