Abstract

The development of storage tissues in seeds (endosperm or cotyledons) involves the multiplication, enlargement, and differentiation of cells with consequent accumulation of different storage products. The biosynthesis of carbohydrates, fats, or other nonprotein components of cells involves a complex regulatory network with a large number of genes. In contrast, storage proteins — such as the maize zeins, the vicilins and legumins of pea, the hordeins of barley, the wheat gliadins – are directly encoded by single genes. At well-defined times, not only the proteins but also the corresponding primary transcripts and mRNAs are present in high concentrations — up to 50% of the cellular RNA — and can be isolated, analyzed, and used for the gene analysis.
