Immunological Intervention with Chimeric CD4 Monoclonal Antibodies

Abstract

The idea of turning the weaponry of the immune system against itself is attributed to Eli Metchnikoff, who used a rabbit antiserum against primitive phagocytes for modulating their function. Today, the great efforts of international leukocyte differentiation workshops charting the unknown territory of the human immune system on the molecular level with the help of the murine immune system have provided hundreds of murine monoclonal antibodies recognizing defined targets on the formerly white map of the human lymphoid cell surface. Rather unexpectedly, monoclonal antibodies have also turned out to be the key to the genes encoding many of the lymphocyte surface molecules which are critical not only for initiation of an adaptive immune response but also for maintenance of memory and induction of tolerance. Some of these molecules are also used for navigational purposes by the mobile lymphocytes and are needed for their functioning as effector cells at appropriate targets.
