Cytochrome P450 Enzymes as Autoantigens in Autoimmune Hepatitis

Abstract

Autoantibodies against several subcellular components of liver tissue are associated with inflammatory liver diseases [1]. While antimitochondrial antibodies are associated with primary biliary cirrhosis (PBC), antinuclear antibodies, liver-kidney-microsomal (LKM) antibodies, and antibodies against soluble liver antigen characterize three different subgroups of idiopathic autoimmune chronic active hepatitis (CAH) (Table 1) [2]. The etiology of these serologically distinct subgroups of CAH is unknown. A loss of tolerance against liver tissue is regarded as the main pathogenetic mechanism. In contrast to virus-induced liver diseases, these autoimmune variants of CAH respond well to immunosuppressive treatment. Furthermore, these disease associated autoantibodies may give further insights into pathogenesis and may provide clues to their etiology. In recent years the identification and characterization of hepatocellular autoantigens has made significant progress related to the development of new techniques, including molecular cloning.
