Zusammenfassung
Die Bösartigkeit solider Tumoren beinhaltet ihre Fähigkeit zur Invasion und Metastasierung. Neuere Ergebnisse der grundlagenorientierten und klinischen Forschung zeigen, daß die Tumorzelle selbst über ein großes Arsenal an Substanzen verfügt, um die Strukturen ihrer unmittelbaren Umgebung aufzubauen und auch wieder aufzulösen. Damit ist es ihr möglich, z.B. über den Abbau der extrazellulären Matrix und der Basalmembran, sich in das sie umgebende Gewebe auszubreiten und in Gefäße einzubrechen. In entsprechender Weise erfolgt durch verschleppte Tumorzellen bzw. Zellembolien nach Adhäsion im Gefäßbereich von Lunge, Leber, Knochen oder anderen Organen die Penetration der Gefäßwand und die Invasion des darunterliegenden Gewebes. Da die Struktur der zu durchdringenden Gewebe vorwiegend von Proteinen, Proteoglykanen und Kollagen gebildet wird, handelt es sich bei den Substanzen, die die Tumorzelle zur Invasion und Metastasierung benutzt, vorwiegend um Proteasen. In besonderer Weise sind es hierbei Faktoren der Blutgerinnung und der Fibrinolyse, die über den Auf- und Abbau einzelner Komponenten der Tumor-Matrix an diesen Prozessen direkt und über die Aktivierung anderer Proteasesysteme indirekt beteiligt sind.
