Zusammenfassung
Das Wachstum maligner Tumore ist einerseits durch Proliferation, andererseits durch die Fähigkeit der Tumorzellen zur Invasion in die Umgebung gekennzeichnet. Während die Regulation der Proliferation sowie ihre biologische und prognostische Bedeutung schon breit untersucht wurden, rückt die Tumorinvasion erst in den letzten Jahren in den Mittelpunkt der Tumorforschung. Heute steht mit verschiedenen Meßmethoden der Tumorzellmotilität in vitro — wie z. B. dem radiären Migrationsassay — und in vitro-Systemen zur Evaluierung der Tumorzell-Invasivität — wie z. B. Konfrontationskulturen — ein brauchbares Rüstzeug für die Untersuchung der Invasivität sowie deren pharmakologische Beeinflußbarkeit zur Verfügung. Nachdem alle diese Methoden jedoch die komplexe in-vivo-Situation nur bedingt berücksichtigen können, tritt als neue Entwicklung die morphologische Forschung mittels Computer-Simulationen und quantitativer Bildanalyse hinzu.
Summary
Tumor growth depends on tumor cell proliferation on the one hand and on tumor cell invasion into the surrounding tissue on the other. Whereas tumor cell proliferation has been extensively investigated for several decades, the mechanisms of tumor invasion have only attracted broad scientific interest during the last years. In vitro tumor cell motility and invasion can now be assessed by a variety of methods, and quantitative data on invasiveness and inhibition of invasion by antiinvasive compounds can be obtained. The radial migration assay and the embryonic chick heart confrontation assay may serve as examples. Recently, a morphological method using image analysis and computer simulations has been additionally introduced for the evaluation of human tumors in vivo.
