Vergleich zwischen Naevuszellnaevi und dysplastischem Naevus mittels DNA-Histogramm-Analyse

Zusammenfassung

Die Autoren führten eine DNA-Histogramm-Analyse von 30 Patienten an ständig lichtexponierten und auch von 30 Patienten an lichtgeschützten Körperregionen, im Vergleich zu normalen Melanozyten und Melanomzellen an Feulgen-gefärbten 6-Mikron-Paraffin-Schnitten durch. Die Analyse erfolgte mittels Barr-Stround-Mikrodensitometer. Drei Histogramm-Typen konnten unterschieden werden: Typ I: euploid DNA-Gehalt mit einem Peak, Typ II: mittlerer DNA-Gehalt um 2c mit zwei Peaks, Typ III: polyploid Zellkerncharakter mit verstärkter Zellproliferation. Dem Typ-I-Histogramm entsprechen 75% der Compound und korialen Naevi. Im Gegensatz dazu wiesen 75% der dysplastischen Naevi den Typ-III-Histogramm auf.

Summary

DNA histogram analysis was undertaken in light-exposed and unexposed skin samples from 30 patients in each case. Normal melanocytes and melanoma cells were used for comparison. Sections of 6 nm diameter from paraffin-embedded material was stained using the Feulgen method and analyzed using a Barr and Stroud microdensitometer. Histograms could be grouped into three categories: type I cells exhibited euploid DNA content with a single peak, and in type II two peaks were seen with a near-diploid DNA content. In type III the number of polyploid cells was elevated, associated with increased proliferation. The type I histogram was most frequent (75%) in compound and dermal nevi; the type III histogram occurred in 75% of dysplastic nevi.
