With the advent of molecular biology, our knowledge on the mechanisms of calcium signalling and signal transduction increased enormously. In this chapter, we highlight some of the spectacular advances and new approaches regarding excitation-contraction coupling in cardiac and skeletal muscle and the mechanisms by which the calcium signal elicits a contractile response and determines contractility.
