Zusammenfassung
Die ersten Beobachtungen über antidepressive WirkungScigenschaften von Pharmaka gehen in die Mitte der 50er Jahre zurück (Kuhn 1957; Pletscher et al. 1960). Ursprünglich als Antihistaminika entwickelte trizyklische IminodibenzylSubstanzen, aber auch Tuberkulostatika zeigten als „Nebenwirkungen“depressionslösende Eigenschaften. Diese klinischen Beobachtungen waren zugleich Ausgangspunkt für eine neue wissenschaftliche Disziplin, die Psychopharmakologie. Klinik und Grundlagenforschung suchten gemeinsam nach Mechanismen von antidepressiven Wirkungen. Mit der Aufklärung der Pharmakawirkungsme-chanismen wurden 2 Ziele angestrebt. Die Entwicklung neuer, möglichst spezifisch angreifender Medikamente und die Entdeckung pathobioche-mischer Strukturen als mögliche Ursachen für depressives Geschehen.
