Zusammenfassung
Noch vor wenigen Jahren wurden pharmakokinetische oder toxikokine- tische Probleme (Pharmaka und Schadstoffe) methodologisch und methodisch einheitlich innerhalb des klassischen Konzeptes der Kompartiment- Modelle behandelt. Nun wird mit der physiologisch orientierten Modellierung (physiologically based pharmacokinetic modeling PB-PK) (/1–8/) eine Linie verfolgt, die ihren Ausgangspunkt in der Pharmakologie (Anästhesiologie) hat, aber eben besonders auf Anforderungen bei der Modellierung (arbeits-)toxikologisch interessierender Biosysteme (Säugetierorganismus als Vesuchstier, Mensch) antwortet. Es wird erwartet, auf diese Weise die Interpretation von Daten vor allem der Toxizitäts- testung (Dosis-Wirkungs-Beziehung) und des Biomonitoring (Beziehng zwischen äußerer Exposition und innerer Belastung/Dosis) zu verbessern. Der auf diese Weise mögliche Zugang zur ‘inneren Exposition’ (target tissue dose) als Ausage über die Konzentration an Schadstoff oder dessen Biotransformationsprodukt in einem bestimmten, nicht mehr nur phänomenologisch als Kompartiment beschriebenen Organ oder Indikatormedium (Blut, Urin) muß als eine wesentliche Voraussetzung dafür angesehen werden.
