Dam Methyltransferase in Escherichia coli: Effects of Different Enzymatic Levels on DNA Replication and Cell Growth

Abstract

A certain level of dam methyltransferase is required for the cells to initiate simultaneously all chromosomal origins of replication. The dam gene of phage T4 was found to substitute for the chromosomal dam gene in this function, strongly suggesting that the effect is mediated by GATC methylation. A sudden overproduction of dam methyltransferase resulted in drastic reductions in the synthesis of RNA, protein, and DNA, by a mechanism that is yet unknown.
