Zusammenfassung
Im Jahre 1988 wurde das Hepatitis-C-Virus (HCV), der häufigste Erreger der Posttransfusionshepatitis (PTH), erstmalig mit molekularbiologischen Methoden charakterisiert (Choo et al. 1989). Das Virus ist ca. 50–60 nm groß, besitzt eine Lipidhülle und wird der Familie der Flaviviridae oder Pestiviridae zugeordnet (Tabelle 1). Inzwischen ist das gesamte Genom des HCV von Isolaten aus den USA (Houghton et al. 1988), Japan (Okamoto et al. 1990; Takeuchi et al. 1990; Takamizawa et al. 1991) und in Teilen aus Europa (Schreier et al. 1991; Fuchs et al. 1991) sequenziert worden. Das Genom hat eine Länge von ca. 9400 Nukleotiden, eine Plusstrangpolarität und einen durchgehenden offenen Leserahmen, von dem ein großes Polyprotein mit einer Länge von 3011 Aminosäuren abgelesen wird, das posttranslationell in die Struktur- und Nicht-Strukturproteine gespalten wird (Tabelle 1, Abb. 1). HCV-Isolate aus den USA und Japan zeigen hohe Homologie in den Nichtstrukturgenen (Takeuchi et al. 1990; Takamizawa et al. 1991), aber deutliche Unterschiede der Aminosäuren-Sequenz in den Envelopeproteinen (30%). Europäische Isolate zeigen hohe Homologie zu den ersten amerikanischen Isolaten (Prototyp-HCV (pTHCV). Neuere Untersuchungen zeigen, daß auch in Deutschland Isolate vom japanischen Subtyp vorhanden sind (Schreier, unveröffentlichte Ergebnisse).
