Zusammenfassung
Im Enantone Monats-Depot wurde das LHRH-Analogon galenisch so eingebaut, daß eine sichere Depotwirkung über mindestens einen Monat ohne Gefahr einer Kumulierung erfolgt. Dazu wurde der Wirkstoff Leuprorelinacetat in einer Matrix aus dem Copolymer von Glycolsäure und Milchsäure (im Verhältnis 1:3) so eingebettet, daß sog. Sphäroide mit einem Durchmesser von 20 pm entstehen [6]. Dieses Copolymer wird wie chirurgisches Nahtmaterial resorbiert [1, 2, 4, 7, 8]. Das Leuprorelinacetat selbst hat eine um mindestens 50fach gesteigerte Wirksamkeit gegenüber dem natürlich vorkommenden GnRH [3]. Die Möglichkeit, durch die tägliche Einmalgabe von 1 mg Leuprorelinacetat den Serumtestosteronspiegel auf das Kastrationsniveau zu senken, konnte gezeigt werden [5].
