Zusammenfassung
Zusammen mit Parathormon (PTH) ist der biologisch aktive Vitamin D Metabolit 1α,25-Dihydroxyvitamin D3 (1,25(OH)2D3) der wichtigste hormonelle Regulator des Calcium-stoffwechsels im Menschen. Neuere Daten zeigen, daß sich die Wirkungen von 1,25(OH)2D3 nicht nur auf Organe, die an der Regulation der Calciumhomöostase beteiligt sind, beschränken. Es steht jedoch außer Frage, daß die zentrale Rolle von 1,25(OH)2D3 in der Beeinflussung der Funktion der bekannten „klassischen“ Zielorgane Darm, Knochen und Niere zu sehen ist. Im folgenden soll die Wirkweise von 1,25(OH)2D3 auf den Knochen genauer diskutiert werden. Im zweiten Teil des Artikels soll auf neuere Aspekte des Vitamin D Stoffwechsels eingegangen werden, die möglicherweise auch Bedeutung für die Regulation des Knochenstoffwechsels durch 1,25(OH)2D3 haben. Es wird detaillierter erläutert werden, inwieweit Zellen des Knochenmarks und des Knochens in der Lage sind, selbst 1,25(OH)2D3 herzustellen, so daß neben der endokrinen Wirkweise des renal produzierten Hormons auch parakrine Effekte am Knochen zu diskutieren sind.
