Zusammenfassung
In einer offenen Studie wurde die Thrombocyten-Aggregation ex vivo nach einer Einmalgabe von 15 ml Rökan (600 mg) untersucht. Die Aggregation wurde durch Adrenalin, Adenosindiphosphat (ADP), Kollagen und PAF-acether induziert und im thrombocytenreichen Plasma gemessen. Die Proben wurden 1 h vor sowie 2, 4 und 8 h nach Verabreichung von Rökan gewonnen. Die Thrombocyten-Aggregation war bei allen PAF-Konzentrationen, bei 1 µM ADP und bei 1 µM Adrenalin reduziert. Die deutlichste Abnahme der Aggregation wurde bei 75 nM PAF-acether 4 h nach der Medikation (p < 0,05) sowie bei 300 nM PAF-acether 4 h (p < 0,01) und 8 h (p < 0,05) nach der Medikation beobachtet. Gerinnungsstatus, Blutungszeit, Blutdruck und Herzfrequenz wurden nicht modifiziert.
