Zusammenfassung
Nach der von Martin postulierten Theorie (Gilbert u. Martin 1976; Martin et al. 1976) liegt der Wirkung der Opiate eine Opiat-Rezeptor-Bindung zugrunde. Diese hängt zum einen von der Affinität des Opiats zum Rezeptor und zum anderen von der intrinsischen Aktivität (Latasch u. Christ 1986) ab, der Fähigkeit, durch die Bindung des Opiats mit dem Rezeptor einen bestimmten Effekt auszulösen. Im Gegensatz zu den Agonisten mit einer Affinität und einer intrinsischen Aktivität haben kompetitive Antagonisten trotz Affinität zum Rezeptor keine intrinsische Aktivität (Latasch u. Christ 1986). Zwischen diesen beiden Gegensätzen liegen die partiellen Agonisten, die eine geringere intrinsische Aktivität haben als die reinen Agonisten.
