Part of the book series: Handbook of Experimental Pharmacology ((HEP,volume 93 / 1))

Abstract

The routine methods employed in the toxicity testing of pharmaceuticals are described and discussed in many textbooks and other publications. Nevertheless, this contribution commences with a brief account of the current methods and principles in order to provide a background for the discussion of a number of findings attributed to an exaggeration of the pharmacological activity of antihypertensive agents during toxicity studies. These findings are either reported in the scientific literature or, in some instances, were made in our own laboratories. On rare occasions it has been necessary to refer to the Federal Drug Administration’s (FDA) Summary basis of approval, reflecting the problem of the pharmaceutical industry’s general diffidence towards the open publication of toxicological data.
