Abstract

A modular sequencer for peptides and proteins has been developed which controls the complete Edman degradation, including isomerization (Wittmann-Liebold et al 1976) and identification of the released amino-acid derivatives by online HPLC (Wittmann-Liebold and Ashman 1985). Because of the modular design, individual units can be exchanged easily, thus enabling the user to introduce new chemistries for sequencing and to design programs. All reagents and solvents are delivered as liquids. A new cross-flow reaction chamber with a very small inner volume (14 μl) is employed. Polyvinylidene difluoride, PVDF, (Madsudaira 1987) or glass fiber support can be used.
