Zusammenfassung

Die isoelektrische Fokussierung (IEF) ist ein Elektrophoreseverfahren in einem pH-Gradienten. Hierbei werden amphotere Makromoleküle wie z. B. Peptide, Proteine, Enzyme und Hormone nach ihren isoelektrischen Punkten (pI) aufgetrennt. Die Bestimmung der Ladungsheterogenität ist ein empfindlicher Nachweis zur Unterscheidung sehr ähnlicher Moleküle. Da die getrennten Fraktionen an ihren isoelektrischen Punkten konzentriert (fokussiert) werden und nicht der Diffusion unterliegen, besitzt die Elektrofokussierung ein äußerst hohes Auflösungsvermögen. Entsprechend zahlreich sind daher die Anwendungsgebiete [1, 2].
