Zusammenfassung
Im Jahre 1676 beobachtete Dodart, daß es bei Vergiftungen mit dem auf Getreideähren schmarotzenden Pilz Claviceps purpurea, dem Mutterkorn, auch zu einer Laktationshemmung kommt. Shelesnyak entdeckte 1954, daß Mutterkomalkaloide bei der Ratte die Implantation der befruchteten Eizellen in die Uterusschleimhaut verhindern. Er führte seine Beobachtungen auf eine Hemmung der Prolaktinsekretion durch diese Substanz zurück, da Prolaktin bei der Ratte in der Frühschwangerschaft als luteotropes Hormon wirksam ist (Althoff et al. 1982). Anfang der 60er Jahre wurden diese Befunde von Baseler Wissenschaftlern wieder aufgegriffen und der Nachweis erbracht, daß das 1965 erstmals synthetisierte Mutterkomalkaloid 2-Brom-α-ergocryptin (Bromocriptin) ein selektiver Hemmstoff der Prolaktinsekretion ist. Zu diesem Zeitpunkt war noch keineswegs geklärt, ob es das Prolaktin beim Menschen als Hypophysenhormon überhaupt gäbe. Dieser Nachweis gelang erst 1970. Parallel dazu konnte sehr früh schon gezeigt werden, daß Bromocriptin die Prolaktinfreisetzung durch eine direkte Wirkung auf die Dopaminrezeptoren in der Hypophyse blockiert. Es gelang der Nachweis, daß Bromocriptin nicht nur die basale, sondern auch die durch physiologische oder pathologische Mechanismen gesteigerte Prolaktinsekretion vermindert. Die Namen von Wissenschaftlern wie Flückiger, Frantz, Kleinberg, Freisen, Lutterbeck, Besser, del Pozo, Hoekfelt, Fuxe und Pasteels sind eng mit der Entdeckungsgeschichte sowohl von Bromocriptin als auch von Prolaktin selbst verbunden (Thomer et al. 1980; Althoff et al. 1982).
