Abstract

Many details of peripheral cholinergic systems have long been part of classical neuroanatomy. The main challenge for some time has been to understand the distribution of cholinergic pathways in the central nervous system. Considerable progress has been made recently, partly from the development and application of the biochemical techniques discussed in the previous chapter but more from histochemical studies which have involved either localization of choline acetyltransferase (ChAT) or of acetylcholinesterase (AChE) following administration of the irreversible cholinesterase inhibitor diisopropylphosphorofluoridate (DFP). Ancillary histological techniques include immunohistochemistry for acetylcholine (ACh), autoradiographic localization of axonally transported radioactive choline, autoradiographic localization of receptors using labelled ligands presumed to be specific for ACh receptors, and double-labelling methods aimed at identifying cholinergic projections or synaptic connections between cholinergic and other neuronal systems. The immunohistochemical localization of Chol-1, a membrane ganglioside apparently specific to cholinergic nerve endings (Richardson et al. 1982; Chaps. 12, 15, this volume), and autoradiography for [3H]hemicholinium-3 ([3H] HC-3), which is bound specifically to the high-affinity choline uptake sites characteristic of cholinergic synapses (Vickroy et al. 1985), offer promise for the future but have not yet been widely applied. Cloning of cRNA for ChAT (Strauss et al. 1985) may open up another avenue for investigating ACh cells.
