Abstract
To maintain germplasm of potato, the ‘seeds’ (tubers) have to be grown and multiplied in nurseries every year. Numerous clones of potato are being maintained at the International Potato Centre, Lima by this method, which is not only time-consuming, labour-intensive, expensive and cumbersome, but above all, the material is exposed to the hazards of pests and diseases, and may result in the total loss of germplasm. Thus, the problem of the conservation of genetic resources of vegetatively propagated plants has encouraged attempts to develop unconventional methods. As an alternative, meristem cultures have been employed for storage (see Sects. V.2 and V.3, this Vol.). However, this method is effective for short-term maintenance as it involves transfer to fresh media and constant vigilance and control of storage conditions. At present, no method is available by which long-term conservation of germplasm of vegetatively propagated plants can be ensured.
