Abstract
The interest in regenerating potato plants through protoplast culture techniques has grown immensely during the last few years, probably due to the vast opportunities offered by this novel method for crop improvement. As a consequence, many different protocols have been published on the isolation and culture of protoplasts from this species (Butenko et al. 1977; Shepard 1980; Thomas 1981; Bokelmann and Roest 1983). Since none of these methods was suitable for the isolation and further culture of some potato varieties of interest to Swedish potato breeding, we had to make some modifications of previously described protocols, and these modifications are described in this chapter. We could increase the number of growing cell colonies from protoplasts by using activated charcoal in order to adsorb substances present in the culture, which otherwise had a deleterious effect on cell viability. Moreover, diluting the medium or lowering the auxin concentration had a promotive effect on plating efficiency.
