Abstract
Identification of domains involved in assembly of the Na,K-ATPase α- and β- subunits is a major focus of our lab. These studies can lead to an understanding of the mechanisms controlling subunit assembly and may provide information about the membrane topology of the α-subunit and folding properties of the β-subunit extracellular domains. Our approach has taken advantage of the unique properties of several monoclonal antibodies that recognize the chicken β1 -subunit. Expression of chimeric subunits and/or subunits lacking portions of the polypeptide chains, followed by immune precipitations with these monoclonal antibodies, has allowed us to identify minimal regions within both the α- and β-subunit amino acid sequences that are sufficient for assembly.
