Growth Inhibitors for Mammary Epithelial Cells

Abstract

In general, we know more about what stimulates the growth of organs than about what regulates their growth. This is particularly true for the mammary gland, in which the systemic mitogenes, the mammogenic hormones, which drive growth have been examined by generations of endocrinologists. However, we do not know which factors or hormones define the final size of a mammary gland even in continued presence of powerful mitogenes. Nor do we know which factors define the typical pattern of ducts and alveoli during the complex process of morphogenesis of the mammary gland during pregnancy and lactation. The mammary gland is a special object for examining processes such as growth and differentiation for several reasons. (1) Breast cancer is one of the leading illnesses during the lifetime of a woman and therefore it is of interest to know more about growth regulation of the mammary epithelia. (2) Branching morphogenesis of the mammary gland is reminiscent of the developmental pattern in other structures such as the lung, kidney, exocrine organs, and various organs of the reproductive tract. (3) Most mammary growth occurs after birth and can be easily studied in mouse, which is large enough to work on conveniently and in which the glands are in an easily accessible location. (4) Finally, cell culture systems either for monolayer growing immortalized, transformed or primary mammary epithelial cells (MEC) and whole-mount culture systems for the mammary gland are well developed.
