Abstract

In grasses, somatic cell and tissue cultures in vitro have been initiated from a wide range of expiants, such as embryos, roots, leaf bases, immature inflorescences and meristems (Yamada 1977; Vasil 1987). When the surface-sterilized expiants are placed on appropriate culture media and maintained axenically under suitable growth conditions, competent cells in the expiant may dedifferentiate and divide to produce a proliferating unorganized mass of cells called callus. Initiated calli or calluses may be maintained in vitro by periodic subcultures onto fresh media to establish callus cultures.
