Zusammenfassung

Trotz intensivster Forschungsanstrengungen sind die Mechanismen, die bei der Entstehung von Tumoren eine Rolle spielen, noch immer nicht vollständig geklärt. Unser Verständnis von der Ätiologie maligner Erkrankungen ist jedoch durch die Entdeckung molekularer und zellulärer Veränderungen in Tumoren gewachsen. Inzwischen ist bekannt, daß Tumoren unterschiedliche genetische Alterationen besitzen und im Verlauf des Tumorwachstums es zu weiteren genetischen Umstrukturierungen kommen kann. Nach heutiger Ansicht sind für eine bösartige Entartung von Zellen mehrere solcher genetischer Variationen notwendig. Diese betreffen v.a. Gene, die für das Wachstum verantwortlich sind. Wichtige Komponenten für das Wachstum sind Protoonkogene, Suppressorgene, Zellzyklusproteine und angiogene Faktoren, wobei eine Zelle in der Regel nicht nur auf einen Faktor, sondern auf ein fein abgestimmtes Zusammenspiel verschiedener Faktoren reagiert.
