T-Region Transfer from Agrobacterium Tumefaciens to Plant Cells: Functional Characterization of Border Repeats

Abstract

Oncogenic Agrobacterium tumefaciens strains can transfer a specific part of their large tumor inducing (Ti) plasmid to the plant cell and thereby cause the plant disease called Crown Gall (Smith and Townsend 1907). The transferred DNA (T-DNA) is stably integrated into the genome of the plant cell (Chilton et al 1977; Thomashow et al 1980). Expression of genes present on this T-DNA results in the production of the plant hormones cytokinin and auxin, which leads to tumor formation on a wide range of dicotyledonous plants (Thomashow et al 1984; Inzé et al 1984; Barry et al 1984; Akiyoshi et al 1984; Buchmann et al 1985). Furthermore unusual amino acid derivatives (opines) are synthesized in the tumor cells as a consequence of the expression of certain T-DNA genes. Which opines are synthesized in the tumors is dependent of the particular Ti-plasmid that was present in the tumor inducing A.tumefaciens strain (Bomhoff et al 1976; Guyon et al 1980). Research has focussed on the socalled octopine and nopaline Ti-plasmids.
