Abstract
The adenine nucleotide carrier (AdN carrier) is a membrane-bound protein (minimum Mr ≃ 32,000) which is responsible for the vectorial exchange between mitochondrial ATP and cytosolic ADP through the inner mitochondrial membrane. During the course of oxidative phosphorylation, the ATP synthesized in mitochondria is exported against ADP generated in cytosol by energy-consuming reactions; this asymmetric exchange is governed by the membrane potential generated by mitochondrial respiration. Two specific inhibitors of the AdN carrier, (carboxy) atractyloside (CATR) and bongkrekic acid (BA), have been used extensively to study the mechanism of ADP/ATP transport. The fact that CATR binds to the carrier from the outside of the mitochondrial membrane and BA from the inside points to the asymmetric orientation of the carrier in the membrane (for review, see Vignais et al. 1985).
