Abstract

The various hypotheses concerning the relationship between neuromuscular blocking activity and interonium structures make the study of almost rigid molecules of particular interest. The use of a fairly fixed steroid skeleton to which reactive side chains are attached (Alauddin and Martin-Smith 1962) was an important step forward in the search for useful curarizing drugs and resulted in a very powerful competitive acetylcholine antagonist, pancuronium (Buckett and Bonta 1966; Buckett et al. 1968; Savage et al. 1971). Reports on its use in human practice started with Baird and Reid (1967) and the drug is still the most widely utilized one. In spite of its general sucess, pancuronium is not yet the ideal curare-like agent because of its relatively long duration of action and of its multicausal cardiovascular stimulating property.
