Summary
For successful treatment of deseases multiple dosing is required in most cases. The work deals with the multiple dosing simulation using analog-hybrid computer EAI-580. Through such simulation the influence of the possible practical situations in multiple dosing on drugs plasma levels can be studied such as influence of dose and dosage interval, influence of different initial dose versus loading dose in order to attain steady state in shorter time, influence of the case when the dose (doses) is omited according to the problems of patients compliance, influence of the case when the dosing for acute treatment (higher dose in shorter dosing intervals) is changed to the chronic treatment (lower dose in longer dosing intervals) during the time of observation. All the mentioned cases were studied for the concrete drugs with different pharmacokinetic and pharmacologic properties.
Zusammenfassung
Erfolgreiche Behandlung fordert häufig eine mehrfache Heilmittelanwendung. Diese Arbeit befasst sich mit der Simulation der Mehrfachdosierung auf dem Analog — hybridrechner EAI-5 80, wobei die Einwirkung der Situationen, die bei der mehrfachen Heilmittelanwendung in der Praxis vorkommen könnten, auf das Heilmittelnivo in Plasma, untersucht wurde und zwar: Einfluss der Dose und des Dosenintervals, Einfluss der Verschiedenen Anfangsdosen in der Beziehung auf die nächsten Dosen mit der Absicht der schnellsten Stationierung, Einfluss auf den Fall wo eine (mehrere) Dose (n) ausgelassen wird (werden), Einfluss auf den Fall wo die akute Behandlung (grössere Dosen in kürzeren Intervallen) durch die chronische Behandlung (kleinere Dosen in längeren Intervallen) erstezt wird. Alle erwähnten Fälle wurden auf konkreten Heilmitteln mit verschiedenen pharmakokinetischen und pharmakologischen Eigenschaften untersucht.
