Abstract

The purpose of this chapter is to discuss recent observations made in this and other laboratories that demonstrate voltage-dependent modulation of Ca-channel currents in cardiac cells by dihydropyridine Ca-channel antagonists and agonists. In order to interpret these observations, the effect of membrane potential on the state of the Ca-channel is first discussed. Then, after reviewing the experimental data that show voltage-dependent changes in drug potency, the results are discussed within the framework of the modulated receptor hypothesis of Hille (1977) and Hondeghem and Katzung (1977) that relates drug-receptor interactions to changes in state of a channel.
