Abstract

Spin-labelled probes are now available for the study of volumes, potentials and pH gradients in membrane vesicles (Mehlhorn et al. 1982). These probes cross the membrane but remain for the most part in the aqueous phases. The addition of an impermeable quenching agent causes broadening of the signal from the external Solution. The remaining sharp signal is then a direct measure of the amount of probe within the vesicles. If the probe is uniformly distributed, this gives the internal volume. If the probe is a membranepermeable ion, it will distribute according to the potential. If it is a weak acid or base which is permeable only in the unionized form, it will distribute according to the pH gradient across the membrane.
