Zusammenfassung
Es ist wenig über den Einfluß von Ernährungsfaktoren auf die Depotfettmobilisation im Serum in vivo und in isolierten Adipozyten in vitro bekannt. Wir haben 7 freiwillige Probanden über 4 Wochen kontrolliert mit wöchentlich wechselnden Diäten ernährt und die Fettmobilisation in vivo vor und während körperlicher Belastung (200 Watt, 18 min) und in vitro untersucht.
Die Serumglycerinspiegel während des Fastens und unter fettreicher Kost waren hoch (112 µmol/1 und 89 µmol/1). Während kohlenhydratreicher Kost und bei Prudent-Diet dagegen niedrig (24 µmol/l und 30 µmol/1). Unter körperlicher Belastung stiegen die Werte in allen Fällen auf ungefähr das Dreifache an.
Es bestand eine enge Beziehung zwischen der Fettmobilisation in vivo in Ruhe und der basalen Lipolyse in vitro, sowie der durch körperliche Belastung stimulierten Lipolyse in vivo und der durch Noradrenalin (10−7 mol/1) stimulierten Lipolyse in vitro. Die Ergebnisse lassen vermuten, daß bei der Adaptation des Fettgewebsstoffwechsels an verschiedene Ernährungsformen neben hormonalen Einflüssen auch hormonunabhängige Änderungen des Zellstoffwechsels eine entscheidende Rolle spielen.
Summary
The influence of different diets on the metabolism of adipose tissue in vitro, as well as on lipolysis in vivo is poorely investigated. We, therefore, kept 7 volunteers on 4 different diets, each for one week. At the end of each week we studied serum glycerol levels during exercise (200 watt, 18 min) and the glycerol release of their isolated adipocytes.
Serum glycerol levels were high after one week of a diet rich in fat and after one week of starvation (89 µmol/1 and 112 µmol/1). On the other hand, they were reduced after one week of a diet rich in carbohydrate or the so called “Prudent-Diet” (reduced in fat content, rich in polyunsaturated fatty acids) (24 µmol/1 and 30 µmol/1). During exercise, serum glycerol increased approximately 3 fold in all conditions mentioned.
Lipolysis in vivo was closely related to the glycerol release of isolated adipocytes in vitro. Serum glycerol levels measured during rest in vivo showed the same response as unstimulated lipolysis of the isolated adipocytes in vitro. A similar pattern was observed when comparing stimulated lipolysis due to physical exercise to stimulation of isolated adipocytes with norepinephrine (10−7 mol/1) in vitro.
