Abstract

The separation of amino acids was the first completely automated chromatographic process with post-column derivatization. The separation was carried out on a cation exchanger and the detection in the eluate after separation was performed by means of ninhydrin1). This method has had its place for more than 30 years in every protein laboratory. Amino acid analysis has taken a further step forward with the appearance of HPLC and the requirement for more rapid and sensitive separation and detection techniques. At first the classical method was merely adapted to the conditions of HPLC by modifying the separation resins and changing and improving the detection methods, more recently the whole area of amino acid analysis has been optimized with regard to the separation system, derivatization before or after separation, improvement of the sensitivity of detection and including automation of the individual steps.
