Application of HPLC to the Separation of Lipids

Abstract

It is comparatively easy to write about “separations” of lipids because, in the field of lipid HPLC, separation techniques as such have made far more progress than detector developments. Today we can separate most of the mixtures that we want to separate, but all too often we cannot detect the separated peaks, or at least we cannot quantitate them. It must be kept in mind, therefore, that in a lot of published lipid work nowadays the separation modes used were chosen primarily with an eye on the detector available — and not because that particular separation mode is the best one to separate the molecules or groups of molecules one wants to separate. Thus, often a gradient would be best but cannot be used because of an RI detector; benzene or toluene would be best for the separation on a silver nitrate column but cannot be used because one needs detection via short-wave UV, or a “group” separation and quantitation would be best but cannot be used because there is no detector with equal sensitivity for saturated and unsaturated molecules present together in one peak.
