Immunocytochemical Characterization of Monoclonal Anti-Leucocyte Antibodies

Abstract

We have previously reported the value of immunohistological screening on tissue sections, both as a rapid method of analysing the reactivity patterns of monoclonal antibodies against individual antigens and as a means of identifying new monoclonal antibodies which would be difficult to detect by conventional screening methods [4]. For the First International Workshop on Human Leucocyte Differentiation Antigens we have screened all the antibodies received in each of the three panels (T, B and macrophage) on a variety of acetone-fixed cryostat tissue sections to establish their pattern of reactivity, to detect any tissue cross-reactions, and to compare the intensity of reaction between similar antibodies. All the antibodies were also screened on paraffin-embedded sections of lymphoid tissue in order to evaluate their possible role in routine histopathological diagnosis.
