Zusammenfassung
Adrenozeptoren sind ursprünglich in die Pharmakologie des sympathischen Nervensystems eingeführt worden, um die quantitativen Wirkungsunterschiede der Katechola-mine Noradrenalin und Adrenalin zu erklären. Ihre einzige physiologische Bedeutung wurde zunächst darin gesehen, daß sie als Ausgangspunkte für die Wirkungen des adrenergen Transmitters Noradrenalin und des Nebennierenmarkhormons Adrenalin dienten. Die ersten Hinweise darauf, daß die Katecholamine ihre Wirkungen über verschiedene Rezeptoren vermitteln, finden sich bereits in den klassischen Arbeiten von Dale (1913), der zeigen konnte (Abb. 1), daß an der spinalisierten Katze die pressorische Wirkung des
Adrenalins nach Vorbehandlung mit Ergotoxin in eine depressorische umgewandelt werden konnte (Adrenalinumkehr) und daraus folgerte, daß sich die Wirkung des Adrenalins funktionell aus einer „exzitatorischen“ und „inhibitorischen“ Komponente zusammensetzt. Den sich daraus ergebenden Schluß allerdings, daß Adrenalin diese verschiedenen Wirkungen durch Stimulation verschiedener Rezeptoren vermittelt, zog erst 40 Jahre später Ahlquist, der 1948 das Postulat von der Existenz verschiedener Adreno-zeptoren in die Pharmakologie einführte. Den experimentellen Nachweis dieser verschiedenen Rezeptoren führte Ahlquist mit den klassischen pharmakologischen Methoden, die noch heute eine der Möglichkeiten für die Differenzierung verschiedener Rezeptoren darstellen: Man mißt an einem definierten Organsystem — in vitro oder in vivo — den Effekt verschiedener Agonisten und stellt dann dafür eine Wirksamkeitsreihenfolge auf.
