Abstract

Apart from the impact of cytokinins on the mitotic activity of plant cells, the requirement of these hormones for chloroplast differentiation is one of the most striking subcellular effects (see Parthier, 1979). In spite of some useful contributions, the intimate nature of the molecular pathway(s) involving cytokinins in the development of plastids is still a matter of speculation. Tissue cultures (Stetler and Laetsch, 1965) and cell suspensions (Lescure, 1973) have been recognized as favorable biological systems to study this phenomenon, keeping in mind the specificity of growth and differentiation status of in vitro cultures compared with intact plants. Some years ago, we undertook to study tobacco cell suspensions in order to obtain stepwise answers to the questions raised by this aspect of cytokinin activities:

  1. a)

    Is it possible to maintain tissue or cell suspension cultures with fully functional chloroplasts?

  2. b)

    How could multiplication and differentiation of plastids be controlled in plant cell suspension cultures, apart from the light requirement?

  3. c)

    Through which molecular activity do cytokinins promote chloroplast differentiation in cell cultures?
