Abstract
The usefulness of vitamin A in the treatment of skin diseases has been limited by the inconsistency of the therapeutical response and by the toxicity at pharmacological concentrations. The search of more suitable compounds has lead to the synthesis of structural analogs of retinol or retinoic acid with increased effectiveness and specificity, combined with lower levels of toxicity. Since 1977 the oral retinoic acid derivative ethyl all-trans-9-(4-methoxy-2,3,6-trimethylphenyl)-3,7-dimethyl-2,4,6,8-nonatetraenoate = Ro 10-9359, has been investigated by us in several dermatoses such as psoriasis, ichthyosis, Darier’s disease, acne, keratodermias, erythrokeratodermia variabilis, pityriasis rubra pilaris, leukoplakia, lichen planus, hypertrophic naevus, monilethrix, congenital pachyonychia, basal cell naevus syndrome and lichen sclerosus et atrophicus vulvae. Based on the excellent results obtained in nearly all disorders of keratinization, the present study is focused on two dermatological conditions — lichen planus and leukoplakia — the treatment of which has been disappointing so far.
