Summary
Using experimental animals it was shown that the water-soluble hydroxyethyl-substituted rutosides [i.e. 3’,4’,7-tri-O-(β-hydroxyethyl)-rutoside and tetra-O-(β-hydroxyethyl)-rutoside] are resorbed to a great extent by the small intestine of the rat. The rutosides were analysed by thin-layer chromatography in the portal and the arterial blood. During the resorption of 3’,4’,7-tri-O-(β-hydroxyethyl)-rutoside, additionally a metabolite was found that was obviously formed in the animal liver. The uptake of the rutosides by the isolated perfused rat liver was shown to be a function of the degree of hydroxyethyl substitution of the hydroxy groups. Tetra-O-(β-hydroxyethyl)-rutoside was found to be eliminated from the blood most slowly; however, 7-mono-O-(β-hydroxyethyl)-rutoside was taken up most rapidly. Free hydroxyl groups are glucuronidated in the liver, as was shown in the case of 3’,4’,7-tri-O-(β-hydroxyethyl)-rutoside.
On the other hand 7-mono-O-(β-hydroxyethyl)-rutoside is split to a nonfluorescent product during passage through the liver. This seems to be the consequence of missing substitution in the side ring. This substitution protects against ring fission.
Rutosides (Venoruton®, injectable) were given intravenously to human volunteers in different doses by a 6 h infusion. A small part of these rutosides was found in the urine of the volunteers, especially when low doses were given. Following the oral administration of 14C-labeled hydroxyethylrutosides only a small part of these substances were found in the urine. These results are not conclusive for a low intestinal resorption rate of the rutosides if the low excretion following intravenous administration is considered.
Zusammenfassung
Im Tierversuch wurde festgestellt, daß die gut wasserlöslichen hydroxyethylsubstituierten Rutoside 3’.4’.7-Tri-O-(β-hydroxyethyl)-rutosid1 und Tetra-O-(β-hydroxyethyl) rutosid1 aus dem perfundierten Dünndarm der Ratte in erheblichem Umfang resorbiert werden können. Der Nachweis der Substanzen erfolgte mittels Dünnschichtchromatographie von Proben aus dem Pfortaderblut und aus dem arteriellen Blut. Während der Resorption von 3’.4’.7-Tri-O-(β-hydroxyethyl)-rutosid wurde ein Metabolit gefunden, der offenbar in der Leber gebildet worden war. Bei Untersuchungen mit der isoliert perfundierten Rattenleber wurde festgestellt, daß die Aufnahme durch das Organ mit zunehmender Substitution abnimmt. Tetra-O-(β-hydroxyethyl)-rutosid wird am langsamsten, 7-Mono-O-(β-hydroxyethyl)-rutosid am raschesten aus dem Blut eliminiert. Freie Hydroxylgruppen von Rutosiden können in der Leber Glucuronidbildung eingehen, wie am Beispiel des 3’.4’.7-Tri-O-(β-hydroxyethyl)-rutosids gezeigt wurde. 7-Mono-O-(β-hydroxyethyl)-rutosid wird bei der Leberpassage zu einem nicht mehr fluoreszierenden Spaltprodukt abgebaut. Dies wird auf das Fehlen von Substituenten im Seitenring zurückgeführt.
Bei freiwilligen Versuchspersonen wurden Rutoside (Venoruton® injectabile) in verschiedener Dosierung bei einer sechsstündigen Dauerinfusion parenteral verabreicht. Nur ein verhältnismäßig geringer Teil der intravenös applizierten Rutoside wurde im Harn der Probanden aufgefunden, insbesondere bei einer geringen Gesamtdosis. Auch bei oraler Applikation von 14C-markierten Hydroxyethylrutosiden wurde bei Versuchspersonen nur ein Bruchteil der markierten Substanzen im Harn wiedergefunden. Dieser Befund ist im Zusammenhang mit der verhältnismäßig geringen Ausscheidung von Rutosiden bei parenteraler Applikation nicht unbedingt gleichbedeutend mit einer geringen intestinalen Resorptionsrate.
