Intravascular administration of ethoxysclerol, thiopental, and carrageenin in the cat’s perfused hindleg at constant flow (chloraloseanesthesia) increased the capillary permeability for albumin. Whether a pretreatment with O-(β-hydroxyethyl)-rutosides (HR) inhibits and/or suppresses these edematous effects was studied. In addition to parameters of blood circulation (blood pressure, arterial perfusion pressure, isogravimetric capillary pressure, venous volume) and the transcapillary fluid exchange, the content of plasma albumin and water in the edematous hindleg was determined and compared to that of the untreated hindleg. HR (50 mg/kg i.v.) inhibited the edema induced by ethoxysclerol and thiopental, much moreso in the skin than in the muscle. The difference in effects presumably was caused by the fact that ethoxysclerol and thiopental produced less edema in the skin, and therefore, their effect there was easier to inhibit than in the muscle. The investigations as evidence for a protective antiedematous effect of the HR against carrageenin were discontinued because the intensity of the edematous effect of carrageenin varied extremely even in controls.
Zusammenfassung
Im strömungskonstant perfundierten Hinterlauf der Katze (Chloralosenarkose) wurde die Kapillarpermeabilität für Albumin durch intravasale Applikation von Ethoxysklerol, Thiopental und Carrageenin erhöht. Untersucht wurde, ob eine Vorbehandlung mit O-(β-Hydroxyethyl)-rutosiden1 (HR) diese ödematösen Effekte hemmt bzw. unterdrückt. Neben Kreislaufparametern (Blutdruck, arterieller Perfusionsdruck, isogravimetrischer Kapillardruck, Venenvolumen) und dem transkapillären Flüssigkeitsaustausch wurde der Gehalt an Plasmaeiweiß und Wasser im ödematösen und zum Vergleich im unbehandelten anderen Hinterlauf bestimmt. HR (50 mg/kg, i.v.) hemmte sowohl das Ethoxysklerol- als auch das Thiopentalödem, und zwar in der Haut stärker als in der Muskulatur. Der Wirkungsunterschied beruhte vermutlich darauf, daß Ethoxysklerol und Thiopental in der Haut schwächere Ödeme erzeugten und dort ihre Wirkung deshalb leichter zu hemmen war als in der Muskulatur. Die Untersuchungen zum Nachweis einer protektiv antiödematösen Wirkung des HR gegenüber Carrageenin wurden abgebrochen, weil die Stärke der ödematösen Wirkung des Carrageenins schon in den Kontrollversuchen erheblich schwankte.
