Zusammenfassung
Es ist allgemein bekannt, daß Patienten unterschiedlich auf dasselbe Medikament reagieren. Meistens wird die Dosierung von Arzneimitteln anhand der Parameter Alter, Geschlecht, Körpergewicht oder Oberfläche und Konstitution des Patienten ermittelt. Man trifft jedoch innerhalb von Bevölkerungsgruppen, ja manchmal sogar in einer Sippe, eine spezifische Variabilität in der Reaktionsweise auf Pharmaka, die entweder eine verstärkte oder aufgehobene Wirkung und vor allem neuartige Nebenwirkungen zeigen, deren weitere Analyse auf eine genetische Belastung hinweist. Aus dieser Beobachtung entwickelte sich das noch relative junge Gebiet der Pharmakogenetik. Im allgemeinen erwartet man auf eine Gabe von bestimmten Arzneimitteln oder Chemikalien eine unimodale Variabilität in Form einer Gaußschen Glockenkurve. Kömmt es aber plötzlich zu einer bimodalen Verteilung der Reaktionen auf ein Arzneimittel, so spricht dieses für genetisch bedingte Variationen, die dann durch Familien-Untersuchungen erhärtet werden müssen. Gerade für den Genetiker ist die diskontinuierliche Verteilung von besonderem Interesse (s. Abb. 1).
