Abstract
An important aspect of the study of photosynthesis is the correlation of photochemical and biochemical activities of the chloroplast thylakoid membrane with its structure. Earlier chapters in this volume have considered the structure and topography of the thylakoid membrane of the mature, fully-greened chloroplast in relation to function, and the properties of subchloroplast fragments obtained by disruption of the mature chloroplast with detergents and mechanical methods. Another approach to the study of chloroplast structure and function is to examine the sequence of biochemical and structural changes during the maturation of the chloroplast. Ideally such a study should be carried out on material which is in synchrony. In the case of higher plants, such as Phaseolus, synchrony is approached by growing etiolated seedlings in the dark for a number of days and then transferring them to light. The use of appropriate etiolated plants also has the advantage that sufficient material is available for biochemical analyses.
