Abstract

The investigations on chloroplast fragmentation reported up to the beginning of the last decade were essentially concerned with the isolation of the “photosynthetic unit” as originally defined by Emerson and Arnold (1932). The major interest was the relationship of particle size to photochemical activity, which during this time was solely characterized by the Hill activity. Some of these classical experiments have been discussed by Thomas (1960) and by Clendenning (1960) in Volume V.1 of the Encyclopedia of Plant Physiology. As photosynthesis became understood in terms of two cooperating light reactions, PS I and PS II, studies in many laboratories were concerned with the preparation of subchloroplast particles enriched in these activities. The increasing interest in this topic is documented by the publication of some excellent review articles (Boardman, 1970; Park and Sane, 1971; Brown, 1973; Arntzen and Briantais, 1975) and by a workshop on special problems of chloroplast fragmentation (Jacobi, 1972). Some laboratories succeeded recently in the isolation of highly purified reaction centers, which are discussed in detail by Wessels (this vol. Chap. IV.4).
